I am planning on driving my car which I recently imported from the US down to he US again. At the Canadian customs the lady told me I would need to contact US Customs, make them aware of the travel and send them the title for a "72 hour check". I called the US customs and asked, they had no idea what I was talking about. My car is fully registered in Canada (I have my sticker and AB plates). Has anyone who's imported run into any issues driving back down to the US? Perhaps the advice I was given was for cars that had yet to complete the full registeration process. Any insight would be appriciated ...

I would assume that you drive your car over the border just like any other car that is registered in AB.

When you crossed the boarder the first time, they "exported" your car. Therefore, to the US, your car is foreign and alien. It would be the same as any other Canadian car as far as they are concerned.
