Based on a $250K budget, building at $150/sqft, your house would be 1600 sqft. That is kinda small for a 40 foot lot. You'd probably get a better return subdividing the lot into 3 smaller 30 feet...
40 x 120 is more than enough. Most infill lots are 25x120.
To build an infill, you'd be looking at around $150 per sqft for standard features.
